Blood collection via orbital sinus --- 眼窝取血

单看“眼窝取血”这四个字,就会觉得恐怖了。确实是挺恐怖的,不过其实不是从眼球取的血,是眼窝里的血管,但还是很可怕的。其实取血的方法有很多中(见reference),从眼窝取血只是其中一种,其他的比较普遍的还包括尾巴静脉(tail vein)取血、脚静脉(saphenous vein)取血、心脏取血(cardiac puncture)。

眼窝取血首先要麻醉老鼠。老鼠麻醉了之后就可以开始进行了。如果有助手帮忙就更好,因为这个过程一个人两只手比较难应付。

要准备的东西包括pasteur pipette或者haematocrit tubes(用来取血的),然后还有收集血液样本的小eppendorf tubes,和纱布。

首先把取血的“武器”对准老鼠眼睛的medial canthus,然后呢,在老鼠目睽睽之下,对了,就“插”进去吧。Medial canthus是眼睛的旁边,不是眼球中间噢。当然插进去的时候不能很大力,要慢慢施力,而且轻轻转动武器。目的是要戳破里面的血管,破了之后血就会流出来,就轻轻把武器提出来一点点,这时因为capillary action血液会自动流上pasteur pipette或者haematocrit tubes里。

替小鼠取血有限制,不能取太多,不然它会失血过多。一般我取100-200microlitres已经很足够了。血取得差不多时,就要把武器交给助手,让他负责把武器中的血液放进小瓶子里,他可以用一个pipette bulb把pasteur pipette里的血液弄出来。而我自己呢,就要在这时马上拿着纱布盖住老鼠的眼睛,以止血,这个步骤非常重要。

通常按了一段短时间后就能移开,移开时用纱布让老鼠的眼皮盖着。如果过程做得好,一般不会给老鼠造成太大伤害。但是如果技巧不好,会造成老鼠失明的噢。

如果戳了很久还是没有血出来,可能是位置错了,角度错了,反正一定有东西错了就对。最重要是能很快地针对情况找到对策。因为你不可能一直不停地戳半个小时的。

以前我做这个步骤,是不替老鼠打麻醉针的,就这样用手restraint着老鼠,然后活生生这样插进去。有时候会听到老鼠疼痛而呻吟的叫声,感觉到老鼠在我的紧握之下不停尝试挣扎,弄到我的手都开始抖了,但又要镇定,不然会拖更久。所以眼球取血是我觉得最难受的行为之一,也是让我觉得自己最残忍的行为。当时我常常告诉自己,自己的行为是为了科学,为了工作,不是为了满足自己的欲望。

那时候取了血之后放开老鼠时,它已经快要奄奄一息了,也不怎么会动弹,就躺在那里,很辛苦的样子。可以想象这样的过程有多疼痛。有大概两只老鼠在这样的严重trauma下突然暴毙。我当时心里还蛮难过的。

后来终于跟上司争取到要使用麻醉针才取血。少了老鼠的挣扎和呻吟却不代表心里难受的感觉也会跟着减少。做一个研究员,心理建设还真的很重要的。

所以呢,从眼睛取窝就是这样啦,但这个过程需要不少勇气和坚定的心。有些事情不能失手,也不能重做,眼球取窝就是这样的一种事情。想想看,如果你戳了很久还不成功,老鼠已经承受了那么多的伤害,你还要三番四次尝试,这样是不是对它来说很残酷?

还是那句,胆大心细。

维也纳马车,Fiaker

从高高的Steffl下来之后,我们又重新走在凉凉冷冷的Stephanplatz上。正当我们走到Stephansdom的后面时,一个新奇的景象又吸引了我的注意力。

我有看错吗?我看到马车也!有人在驾的马车噢!

我不知道要怎么问朋友,我该问她为什么有马车,还是,这是马车吗?哈哈。她对眼前的马车似乎视若无睹。我问,“Do you have this everyday?”,一边指着不远处在缓缓走动的马车。她大概对我这样奇怪的问题觉得费解,“Oh..yes, everyday, yes...”。看来她是完全不知道我没看过这样的马车,哈哈。

然后我又喃喃,哇,everyday oh。。。他们有马车走在路上,每天噢。为什么有马车的咧?在我还没来得及给自己想到一个答案的时候,我们沿着Stephansdom的后面走,一转弯,不得了!

“Oh wow...you have so many of them here!” 我真的情不自禁啦,因为我看到一整排的马车停在Stephansdom北塔的旁边。这时我朋友终于注意到我的反应,开始明白我对眼前所看到的景象觉得很新奇。她才开始解释,“Yeah...they are very expensive taxi in Vienna...” 然后讪讪地笑笑。

我也忘记了旁边的Stephansdom了,径自走到最前面的两头白马儿的附近,哇塞,我站在马车的面前也!这种马车我们只有在玩具模型,和古装的英文片才会看到的也!两头白马瞪着圆骨碌的大眼睛望着我这个大惊小怪的游客,哈哈。

我马上要朋友跟我照相,脸上笑容自然是掩饰不了。马儿看到我在旁边,还想凑过头来舔我,我当然是闪啦。它们虽然长得很漂亮,不过噢,是有股马骚味啦,哈哈。

然后我又照了许多照片。每辆马车都装饰得不同,却都很复古。而每辆马车上的马儿也会不同,有些是白马,有些黑马,另外一些褐色的。有些马大,有些马小。有些马头上戴帽子,有些戴着眼罩。还有还有,它们的屁股,都挂着一个好大好大的袋子,呵呵,不用人家解释也明白,是它们的pampers,哈哈。。。那些味道大概也从这个袋子里传出来的吧。

马儿,为什么你要戴口罩的呢?

两头黑马王子,酷!

我问我朋友,为什么它们有些有眼罩的呢?她说,那是因为要避免当它们走在路上时,被车子或别的东西刺激到,然后乱走噢。我很留意地看着眼前的景象。马儿的脚上有大大方方的马蹄。还有一头马儿伸长舌头不懂在干嘛,我朋友知道我兴奋,快点叫我过去拍照,哈哈。

我朋友看到我很兴奋,知道我是第一次看到这样的马车。可是她似乎不怎么喜欢这些马车,她本来就是一个爱好动物的人,她说这样其实对马儿不好。我想想也是,天气这么冷了这些马儿还要在这里等客人,而且,它们的生活应该很无聊吧,每天就是戴着那个眼罩绕市区走来走去。 不过这样华丽的车子,真的能吸引到游客噢。至少能吸引到我!可是吸引归吸引,我们没有去搭这类叫做Fiaker的马车,等哪一天我很有钱的时候再去维也纳,才考虑要不要坐吧。哈哈,20分钟要EUR40噢,不便宜呢!

香港美食:镛记烧鹅

提到香港的烧鹅,不能不想起镛记。在我们还没有去香港之前,有一次在一个电视美食节目中,看到陈启泰介绍镛记的烧鹅,已经看得目不转睛。我们当时说,好,我们去香港也要吃这个!


镛记在中环兰桂坊的整栋大楼,不错吧,卖烧鹅卖到这样。

下面摘自我的旅游日记

晚上8点的中环熙来攘往的,人潮很多。我们在步行前往兰桂坊的途中,就看到了威灵顿街(Wellington Street)上那个偌大的金碧辉煌的招牌----镛记(Yung Kee)酒家。我们打算今晚在镛记打包两只闻名遐迩的烧鹅腿当作晚餐。

我们本来就不打算在这里(兰桂坊) 吃饭或喝酒,于是走了一下,拍了一些照片,就走回去镛记。之前一直顾着拍镛记的照片,都没发觉到镛记的对面也是出名的黄枝记(Wong Chi Gei)。然后,我们推了镛记那扇重甸甸的大门进去。

只见里面坐着一围围的台,空间不会很大,但相信楼上也能坐人的,应该是一整栋建筑物都是镛记的。很多人在忙着吃晚餐。

Mars主动跟服务员说我们要“打包”,他是用广东话讲的。服务员听了,就说,噢,“外卖啊,入边啦”。我们就跟着指示走到里面的一个柜台,原来他们打包要说是外卖,而镛记的外卖已经电脑化了。首先在柜台里下order付钱,然后再拿着条子到厨房等候。

Mars看到价钱,是HKD90一个烧鹅腿。我们没什么时间process这个价钱,虽然觉得有点贵,但因为一心想要吃,也就没有什么考虑的买了。在厨房外面等待我们的烧鹅腿时,有个穿着西装的男生(香港真的随处可见“西装友”,哈哈)也在旁边坐着等。

后来我们就拎着烧鹅,搭地铁回到油麻地。回去酒店之前还去了Wellcome买了一袋橙和一瓶大装的汽水,哈哈。橙蛮便宜的,一大袋才HKD11.80,汽水HKD9.90。


镛记烧鹅加橙和汽水,不错吧?呵呵。

进房间后,已经9点了。我们就马上打开我们的镛记烧鹅。镛记的外卖果然不是盖的,只是盒子就很讲究,尽量避免里面的汁流出来。此外还附送了一小瓶看起来是豆酱的东西,还有一小盒吃起来像酸梅酱的,我们把这些酱都倒进去。



真凭实据,两只烧鹅腿HKD180。

这时离我们打包已经快45分钟一小时了,烧鹅已经不再温热了,不然吃起来一定更美味的。烧鹅的味道像极了烧鸭,我实在分不出来。这烧鹅虽然凉了,但还是蛮好吃的。

吃饱后我们还吃橙。想不到看起来外表平平的橙竟然那么地甜~哈哈~

后记

就这样,我们在香港吃了第一餐晚餐。虽然两只烧鹅腿有点贵(呵呵),但是吃过了之后还是觉得值得啦~加上我们还有饭后水果和汽水,不错吧?我们也是有情趣的人噢,哈哈!

如果有时间又有budget,大可在镛记里面吃一餐美味的烧腊餐,应该是不错的享受来的~

香港兰桂坊

兰桂坊是香港中环有名的酒吧区。正确来说,兰桂坊是几条酒吧云集的巷弄。这里除了有酒吧之外,还有各种不同国家的餐厅,舞厅以及各式消费场所,洋溢特有的欧陆风味和异国情调,被誉为香港特有的文化特区。

交通

兰桂坊最靠近的地铁站是中环地铁站D出口。

当天我们看完了香港夜景,就从太平山顶乘搭15C巴士下山。巴士很快就停在毕打街(Pedder Street),我们也下车了。跟着路旁的旅游牌子,我们过了一些马路,很快就看到了兰桂坊的德已立街(D' Aguilar Street)。

兰桂坊其实不大,就几条简单的街道。街道是有点斜坡的,两旁都是双层楼的店屋,楼下的店都是各式各样的酒吧和餐厅。很多外国人在这里喝酒,有点像新加坡的克拉克码头(Clarke Quay)。

我们本来就不打算在这里吃饭或喝酒,于是走了一下,拍了一些照片,就离开了,因为要张罗我们的晚餐。

写伊眉儿 - Writting Electronic Mail (From Shirley Taylor, Model Business Letters, Emails ... )




伊眉儿(email),对现在企业的影响很大,有时候每天要花上半天的时间回这些email,为了加强自己的email能力, 还特地去买了这本书来看。 以下是我觉得对自己还蛮有用的。。。

The Internet has made it possible for us to communicate with people
from all over the world.
The only way those people can form an opinion on us is by looking at the way we write. Your credibility could be ruined with one swift click of the "send" button.

Do NOT feel obliged to give a detailed reply.
If you are pushed for time and cannot reply immediately, don't feel compelled to do so.
This will only result in a rushed message, perhaps mistakes and perhaps it will not be as detailed or effective as it could be. Instead send a quick note saying you will get bck to the writer soon with a more considered response. This way the writer knows you have received the message and are dealing with it.

Pick up the phone when is needed.

If you have been involved in a prolonged email exchange that lasts for days, wouldn't it be better to pick up the phone ? Email overload is contributing to a decline in oral communication skills.

Creating Electronic Rapport

Studies show that it takes 15 seconds for your customer to judge you when you first meet and greet him or her.
The first impression a customer receives is unfluenced by three things:
- Body language
- tone and voice
- words.
But in email you have only the last 2.

Putting into practice the principles of good customer care is easier in person than on the telephone because a real person can be seen and heart.

It's easier on the telephone than on email becuase even without seeing the person you can hear them and the tone of voice used - and yes, you can hear a smile in a voice.

On email you haven't got either these advantages, so you have to take other steps to try to create electronic rapport with customers as well as collagues.

Lead the reader into the message by backtracking or giving some basic background.

Be warm and friendly in your opening where appropriate. Example,
- Thanks for you lunch last week. It gave us a good opportunity to learn moare about your new project, which sounds very interesting.
- Thanks for calling me today. It made a nice change to speak to a real person for once insttead of always using email.
- I am glad we were able to speack on the telephone this morning. It was good to clarify this issus.
... ... etc.

Try to remember that emotive and sensory words add texture and dimension to the general message of what you are writting.
You owe it to your customers and colleagues to show empathy through email, using language that will help you to form a better bond. Example,
- I'll be pleased to help you sort out this problem.
- I appreaciate your understanding in trying to resolve this issue.
- I see what you mean and can appreaciate your concern

Don't add so much that you come over as too gushy. You need only a few words to really add something extra to your message and show some extra warmth.

Identify with your reade, appreciate their feelings, and use words they will understand, written iin a appropriate tone.

Some other tips:
- Use short sentences and short paragraphs.
The shorter your messsages, the more likey it is that they will be read and understood, but not too short.

- Enumerate with numbers or bullets.
This will add the clarity of your message.

TA: Chart Patterns

A chart pattern is a distinct formation on a stock chart that creates a trading signal, or a sign of future price movements.

Based on the assumption that history tends to repeat itself, the idea is that certain patterns are seen many times, and that these patterns signal a certain high probability move in a stock.

There are two types of patterns within this area of technical analysis, reversal and continuation. A reversal pattern signals that a prior trend will reverse upon completion of the pattern. A continuation pattern, on the other hand, signals that a trend will continue once the pattern is complete.

Below are some common chart patterns:

1. Head and Shoulders
-reversal pattern
-2 versions:
head and shoulders top (formed at the high of an upward movement, signals end of the uptrend)
head and shoulders bottom or inverse head and shoulders (signals a reversal in a downtrend)
-consists of four main parts: two shoulders, a head and a neckline.
-each individual head and shoulder is comprised of a high and a low.
-the neckline is a level of support or resistance.



2. Cup and Handle
-bullish continuation pattern
-in which the upward trend has paused but will continue upward once the pattern is confirmed.
-this price pattern forms what looks like a cup, which is preceded by an upward trend.
-the handle follows the cup formation and is formed by a generally downward/sideways movement.
-once the price movement pushes above the resistance lines formed in the handle, the upward trend can continue.
-wide ranging time frame for this type of pattern, ranging from several months to more than a year.



3. Double Tops and Bottoms
-reversal pattern
-created when a price movement tests support or resistance levels twice and is unable to break through.
-often used to signal intermediate and long-term trend reversals.



4. Triangles
-3 types:
symmetrical triangle, ascending and descending triangle
-last anywhere from a couple of weeks to several months.
-symmetrical triangle: two trendlines converge toward each other, neutral in that a breakout to the upside or downside is a confirmation of a trend in that direction
-ascending triangle: the upper trendline is flat, while the bottom trendline is upward sloping, generally bullish pattern, look for an upside breakout.
-descending triangle, the lower trendline is flat and the upper trendline is descending, generally bearish pattern, look for a downside breakout.



5. Flag and Pennant
-short-term continuation patterns
-formed when there is a sharp price movement followed by a generally sideways price movement.
-then completed upon another sharp price movement in the same direction as the move that started the trend.
-last from one to three weeks.
-difference between a pennant and a flag can be seen in the middle section of the chart pattern.
-in a pennant, the middle section is characterized by converging trendlines, much like what is seen in a symmetrical triangle.
-in a flag, the middle section is characterized by channel pattern , with no convergence between the trendlines.
-in both cases, the trend is expected to continue when the price moves above the upper trendline.



6. Wedge
-either a continuation or reversal pattern.
-similar to a symmetrical triangle except that the wedge pattern slants in an upward or downward direction, while the symmetrical triangle generally shows a sideways movement. -the other difference is that wedges tend to form over longer periods, usually between three and six months.
-at the most basic level, a falling wedge (two lines converge downwards) is bullish and a rising wedge is bearish.



7. Gaps
-an empty space between a trading period and the following trading period
-occurs when there is a large difference in prices between two sequential trading periods (eg. if the trading range in one period is between $25 and $30 and the next trading period opens at $40)
-can be found on bar charts and candlestick charts but will not be found on point and figure or basic line charts.
-show that something of significance has happened in the security, such as a better-than-expected earnings announcement.
-3 main types:
breakaway: forms at the start of a trend
runaway (measuring): forms during the middle of a trend
exhaustion: forms near the end of a trend.

8. Triple Tops and Bottoms
-reversal patterns
-are formed when the price movement tests a level of support or resistance three times and is unable to break through; this signals a reversal of the prior trend.
*Confusion can form with triple tops and bottoms during the formation of the pattern because they can look similar to other chart patterns. After the first two support/resistance tests are formed in the price movement, the pattern will look like a double top or bottom, which could lead a chartist to enter a reversal position too soon.



9. Rounding bottom or saucer bottom
-long term reversal pattern
-a shift from a downward trend to an upward trend.
-last anywhere from several months to several years.
-looks similar to a cup and handle pattern but without the handle.
*The long-term nature of this pattern and the lack of a confirmation trigger, such as the handle in the cup and handle, makes it a difficult pattern to trade.

TA: Chart Types (图表的种类)

有四种普遍被使用的图表:

1. Line Chart

-最基本和简单的
-显示一段时期里每天的闭市价
-不提供详细的价格资料,如每天的最高、最低、开市价格
-不过闭市价本身可以算是一个相当重要的价位(比最高和最低价重要,因此在这里才被应用。)



2. Bar Chart

-比line chart复杂,因为它在每个点上多加了一些重要资料
-由无数垂直bar组成
-每条垂直bar上包含了当日最高、最低和开市价、闭市价
-开市和闭市价是垂直bar两旁的纵横线
-开市价是垂直bar左边的横线
-闭市价是垂直bar右边的横线
-如果开市价比闭市价低,该垂直bar就会涂黑色,代表该股价位上升
-反之,若开市价比闭市价低,该bar就会涂红色
-垂直bar的最高点就是该日最高价,反之,最低点就是最低价



3. Candlestick Charts

-跟bar chart类似,只是呈现方法有点不同
-Candlestick也是由一系列垂直bar所组成,只是它的bar比较宽,呈现开市与闭市价的方法不同
-而且,同样地,candlestick也依赖不同的颜色来显示价位上升或下降(至于什么颜色就看不同的chart analysis,会有分别)
-通常,上升的bar是白色或无色的
-下降的bar是红色或黑色
(不同的颜色要看不同的chart analysis site,务必先搞清楚才研究图表)

B:开市比闭市价高
C:闭市比开市价高,闭市为当日最高价,开市为当日最低价

4. Point and Figure Charts

-比较不普遍
-但历史悠久
-显示价位的移动,但在每个点上不显示时间和交易量
-删除各种“噪音”(noise),即不重要的价位移动
-中和时间对于chart analysis所造成的skewing effect
-由一系列X和O所组成
-X代表上升价位趋势
-O代表下降价位趋势
-图表上每个box代表一个price scale,比如说一个box代表$1



总结

图表是TA的基本工具,因此要了解图表上所确切显示的资料,才能运用图表来进行分析! 这是“工欲善其事,必先利其器”啊!

亿万富翁的神奇公式 



假定有一位年轻人,从现在开始能够定期每年存下1.4万元,如此持续40年;但如果他将每年应存下的钱都能投资到股票或房地产,并获得每年平均20%的投资报酬率,那么40年后,他能积累多少财富?一般人所猜的金额,多落在200万元至800万元之间,顶多猜到1000万元。然而正确的答案却是:1.0281亿,一个令众人惊讶的数字。这个数据是依照财务学计算年金的公式得之,计算公式如下:1.4万(1+20%)40=1.0281亿。  

这个神奇的公式说明,一个25岁的上班族,如果依照这种方式投资到65岁退休时,就能成为亿万富翁了。投资理财没有什么复杂的技巧,最重要的是观念,观念正确就会赢,每一个理财致富的人,只不过养成了一般人不喜欢、且无法做到的习惯而已。  

当然,维持20%的报酬率不容易,单靠股票投资风险很大,有钱的话不妨可以投资房屋地产.
我希望在股市赚了第一桶金后可以投资在房地产,晚年靠租金养老也不错。

诺贝尔基金会

世界闻名的诺贝尔基金会每年发布奖项必须支付高达500万美元的奖金。我们不禁要问:诺贝尔基金会的基金到底有多少?事实上,诺贝尔基金会的成功,除了诺贝尔本人在100年前捐献一笔庞大的基金外,更重要的是归功于诺贝尔基金会的理财有方。诺贝尔基金会成立于1896年,由诺贝尔捐献980万美元。由于该基金会成立的目的是用于支付奖金,管理不允许出现任何的差错。因此,基金会成立初期,其章程中明白地确定基金的投资范围,应限制在安全且固定收益的项目上,如银行存款与公债。这种保本重于报酬率、安全至上的投资原则,的确是稳重的做法。但牺牲报酬率的结果是:随着每年奖金的发放与基金运作的开销,历经50多年后,到1953年该基金会的资产只剩下300多万美元。  

眼见基金的资产将消耗殆尽,诺贝尔基金会的理事们及时觉醒,意识到投资报酬率对财富积累的重要性,于是在1953年做出突破性的改变,更改基金管理章程,将原来只准存放银行与买公债,改变为应以投资股票、房地产为主的理财观。资产管理观念改变后,就此扭转了基金的命运。1993年基金的总资产滚动至2亿多美元。

周末拍拖记(三)

Mars从昨晚到今天
都在忙着configure公司system的东西
搞到晕头转向的样子

出去走走吧
于是我们就出去吃了饭
逛了一下购物中心
很多人啊

差不多要回家的时候
他说,“出来这样走下,心情真的没有那么stress哦~”
